    Procedurally generating surface detail for 3D models using voxel-based cellular automata

    Procedural generation is used extensively in the field of computer graphics to automate content generation and speed up development. One particular area often automated is the generation of additional colour and structural detail for existing 3D models. This empowers artists by providing a tool-set that enhances their existing work-ow and saves time. 3D surface structures are traditionally represented by polygon mesh-based models augmented by 2D mapping techniques. These methods can approximate features, such as caves and overhangs, however they are complex and difficult to modify. As an alternative, a grid of voxels can model 3D shapes and surfaces, similar to how 2D pixels form an image. The regular form of voxel-based models is easier to alter, at the cost of additional computational overhead. One technique for generating and altering voxel content is by using Cellular Automata (CA). CAs are able to produce complex structures from simple rules and also easily map to higher dimensions, such as voxel datasets. However, creating CA rule-sets can be difficult and tedious. This is especially true when creating multidimensional CA. In our work we use a grammar system to create surface detail CA. The grammar we develop is similar to formal grammars used in procedural generation, such as L-systems and shape grammars. Our system is composed of three main sections: a model converter, grammar and CA executor. The model converter changes polygon-mesh models to and from a voxel-based model. The grammar provides a simple language to create CA that can consider 3D neighbourhoods and query parameters, such as colour or structure. Finally, the CA executor interprets the produced grammars into surface-oriented CAs. The final output of this system is a polygon-mesh model, altered by the CA, which is usable for graphics applications. We test the system by replicating a number of CA use-cases with our grammar system. From the results, we conclude that our grammar system is capable of creating a wide range of 3D detail CA. However, the high resolution of resulting meshes and slow processing times make the process more suited to o_-line processing and pre-production

    Megapixel imaging of (micro)nutrients in mature barley grains

    Understanding the accumulation and distribution of essential nutrients in cereals is of primary importance for improving the nutritional quality of this staple food. While recent studies have improved the understanding of micronutrient loading into the barley grain, a detailed characterization of the distribution of micronutrients within the grain is still lacking. High-definition synchrotron X-ray fluorescence was used to investigate the distribution and association of essential elements in barley grain at the micro scale. Micronutrient distribution within the scutellum and the embryo was shown to be highly variable between elements in relation to various morphological features. In the rest of the grain, the distribution of some elements such as Cu and Zn was not limited to the aleurone layer but extended into the endosperm. This pattern of distribution was less marked in the case of Fe and, in particular, Mn. A significant difference in element distribution was also found between the ventral and dorsal part of the grains. The correlation between the elements was not consistent between and within tissues, indicating that the transport and storage of elements is highly regulated. The complexity of the spatial distribution and associations has important implications for improving the nutritional content of cereal crops such as barley

    The role of scaffolding and motivation in CSCL

    Recent findings from research into Computer-Supported Collaborative Learning (CSCL) have indicated that not all learners are able to successfully learn in online collaborative settings. Given that most online settings are characterised by minimal guidance, which require learners to be more autonomous and self-directed, CSCL may provide conditions more conducive to learners comfortable with greater autonomy. Using quasi-experimental research, this paper examines the impact of a redesign of an authentic CSCL environment, based upon principles of Problem-Based Learning, which aimed to provide a more explicit scaffolding of the learning phases for students. It was hypothesised that learners in a redesigned ‘Optima’ environment would reach higher levels of knowledge construction due to clearer scaffolding. Furthermore, it was expected that the redesign would produce a more equal spread in contributions to discourse for learners with different motivational profiles. In a quasi-experimental setting, 143 participants collaborated in an online setting aimed at enhancing their understanding of economics. Using a multi-method approach (Content Analysis, Social Network Analysis, measurement of Academic Motivation), the research results reveal the redesign triggered more equal levels of activity of autonomous and control-oriented learners, but also a decrease in input from the autonomous learners. The main conclusion from this study is that getting the balance between guidance and support right to facilitate both autonomous and control-oriented learners is a delicate complex issue

    Jednym z stosunkowo nowych obszarów badań współczesnej nauki o zarządzaniu jest crowdsourcing oraz zachodzące w nim wirtualnym dzieleniu się wiedzą. Jest ono definiowane jako rozpowszechnienie wiedzy przez społeczność wirtualną, informowanie innych, podawanie jej do wiadomości publicznej, oczekiwanie, że inni tę wiedzę skomentują, rozszerzą i uzupełnią. Takie dzielenie się wiedzą jest szczególnie istotne dla współtworzenia, partycypacji czy uzyskiwania innowacyjnych pomysłów przez organizację. Jednak, pomimo jego pozytywnego wpływu na organizację, dotychczas nie było ono przedmiotem kompleksowych badań. Artykuł przedstawia istniejący dorobek w zakresie sposobów pomiaru społecznościowego dzielenia się wiedzą w ramach crowdsourcingu. W opracowaniu można też znaleźć wyjaśnienia, dlaczego warto badać wirtualne dzielenie się wiedzą.One relatively new area of contemporary science research on management is crowdsourcing and virtual knowledge sharing occurring within it. It is defined as the dissemination of knowledge by a virtual community, informing others, making it public, expecting that others will comment on this knowledge, expand and complete it. Such a sharing of knowledge is particularly important for co-creating, participating, or acquiring innovative ideas by an organization. However, despite its positive impact on the organization, it has not been the subject of comprehensive research so far. This article presents the existing output in the scope of the ways of measuring community knowledge sharing within crowdsourcing.     Guidelines for the use and interpretation of assays for monitoring autophagy (3rd edition)

    In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. For example, a key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process versus those that measure fl ux through the autophagy pathway (i.e., the complete process including the amount and rate of cargo sequestered and degraded). In particular, a block in macroautophagy that results in autophagosome accumulation must be differentiated from stimuli that increase autophagic activity, defi ned as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (inmost higher eukaryotes and some protists such as Dictyostelium ) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the fi eld understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. It is worth emphasizing here that lysosomal digestion is a stage of autophagy and evaluating its competence is a crucial part of the evaluation of autophagic flux, or complete autophagy. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. Along these lines, because of the potential for pleiotropic effects due to blocking autophagy through genetic manipulation it is imperative to delete or knock down more than one autophagy-related gene. In addition, some individual Atg proteins, or groups of proteins, are involved in other cellular pathways so not all Atg proteins can be used as a specific marker for an autophagic process. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field

    Crop biofortification is a sustainable and cost-effective strategy to address malnutrition in developing countries. This review synthesizes the progress toward developing seed micronutrient-dense cereals and legumes cultivars by exploiting natural genetic variation using conventional breeding and/or transgenic technology, and discusses the associated issues to strengthen crop biofortification research and development. Some major QTL for seed iron and zinc, seed phosphorus, and seed phytate in common bean, rice,J;md wheat have been mapped. An iron reductase QTL associated with seed-iron ~QTL is found in common bean where the genes coding for candidate enzymes involved in phytic acid synthesis have also been mapped. Candidate genes for Ipa co segregate with mutant phenotypes identified in rice and soybean. The Gpe-B1 locus in wild emmer wheat accelerates senescence and increases nutrient remobilization from leaves to developing seeds, and another gene named TtNAM-B1 affecting these traits has been cloned.
